No, it isn't the problem. Sendmail doesn't connect to that socket
thus neither knows about it nor cares about it. As for fixing it,
there is nothing to fix. Those are the normal permissions for it. The
idea being that clamdscan users can use clamd without having to have
any special permissions. Of course, having clamd.sock in
/var/spool/MIMEDefang kinda defeats that idea.

You didn't show the permissions on /. Also, what is the version
of MIMEDefang you are using, and what OS are you using? Also, are you
getting these errors continuously or just briefly at startup time?
